The Elementary Teachers Federation of Ontario has filed a complaint with the province's labour board.
The union, which is negotiating a new contract, says the government violated bargaining obligations.
The federation says last month, a memo requiring teachers to conduct mandatory reading screenings twice a year for kids in kindergarten through grade 2 was circulated.
The union says early reading screening is a subject of central bargaining, and the memo violates good faith duties.
The memo says the new requirements must be implemented in alignment with collective agreements.
